Er arbeitete im Bereich der Kolloidchemie und gilt als Entdecker der Liesegangschen Ringe. Im Jahr 1908 ging Liesegang ans Senckenberg-Museum in Frankfurt, wo es ihm am Neurologischen Institut gelang, die feinen Verästelungen der Neuronen im Gehirn sichtbar zu machen. Auf Grund der Bekanntheit seiner wissenschaftlichen Arbeiten wurde er nach dem Ersten Weltkrieg Mitarbeiter am Kaiser-Wilhelm-Institut für Biophysik in Frankfurt am Main, 1937 übernahm er dort das Institut für Kolloidforschung. 1944 wurde das Institut nach Bad Homburg verlegt. Seine Assistentenzeit verbrachte er in Straßburg bei Schmiedeberg und Hofmeister, habilitierte sich daselbst 1897 für physiologische Chemie und war von 1912-18 o. Hon. Professor in Straßburg. 1921 folgte er einem Rufe als o. Professor für physiologische und vegetative Physiologie nach Basel. Von den Arbeiten Spiros, der zu den Führern auf dem Gebiet der Biochemie gehörte, seien die über Mineralstoffwechsel, Eiweißchemie und Gerinnungslehre sowie seine Studien zur physikalischen und Elektrochemie (Quellungsdruck der Kolloide, Pufferung, Ionengleichgewicht, Rolle der Kolloide bei der Transfusion) hervorgehoben. Eingehend beschäftigte er sich auch mit der Physiologie und Pharmakologie der Lymphbildung und der Harnsekretion sowie mit Fragen der Antiseptik. Mit W. Filehne führte er das von F. Stoltz entdeckte Pyramidon in den Arzneischatz ein.
