Professor in Straßburg. 1921 folgte er einem Rufe als o. Professor für physiologische und vegetative Physiologie nach Basel. Von den Arbeiten Spiros, der zu den Führern auf dem Gebiet der Biochemie gehörte, seien die über Mineralstoffwechsel, Eiweißchemie und Gerinnungslehre sowie seine Studien zur physikalischen und Elektrochemie (Quellungsdruck der Kolloide, Pufferung, Ionengleichgewicht, Rolle der Kolloide bei der Transfusion) hervorgehoben. Eingehend beschäftigte er sich auch mit der Physiologie und Pharmakologie der Lymphbildung und der Harnsekretion sowie mit Fragen der Antiseptik. Mit W. Filehne führte er das von F. Stoltz entdeckte Pyramidon in den Arzneischatz ein.

Untersucht außerdem, wie Ernährung und Dosis die Stärke der Glykosurie beeinflussen. Früher Beitrag zur Klärung der Phlorhizin-Glykosurie als Modell des Diabetes mellitus vor der Einführung des Insulins.
