In dieser Arbeit wurde ein systembiologischer Ansatz angewandt, um den zentralen Kohlenstoffmetabolismus von P. aeruginosa zu charakterisieren und Adaptationseigenschaften sowie die Pathogenität des Mikroorganismus umfassend zu untersuchen. Mit Hilfe der 13C-metabolischen Flussanalyse konnten zentrale in vivo-Enzymaktivitäten und Stoffwechselwege des Modellorganismus P. aeruginosa PAO1 aufgeklärt und anschließend die Analysemethode auf 11 uropathogene klinische P. aeruginosa-Isolate aus Harnwegsinfektionen sowie 12 Isolate von Patienten mit einer katheterassoziierten Harnwegsinfektion übertragen werden. In dieser umfassenden Untersuchung wurde ein systemorientierter Ansatz präsentiert, um zielgerichtet Schlüsselreaktionen im zentralen Stoffwechselmetabolismus zu identifizieren, die Targets für zukünftige Vermeidungsstrategien darstellen.

Even though urban and peri-urban agriculture (UPA) is well appreciated by the Food and Agriculture Organi…zation (FAO) and United Nations (UN), it is often criticized due to its intensive mode of production and safety aspects. Only little is known about the UPA production system in the Mumbai Metropolitan Region (MMR). In order to characterize the various UPA production systems across MMR, a baseline survey was conducted in 2010 and further detailed investigations were conducted for general soil characterization and heavy metal signatures.Based on the results of a baseline survey of 165 households, the UPA production systems in the MMR were characterized according to their socio-economic, demographic, migratory and features related to production system. Five major UPA systems were detected: railway gardens, balcony gardens, terrace gardens, farms and ¿Tabelas¿ ¿ enclosures where buffalos and cows are reared for milk. A great diversity of vegetables, fruits, plantation crops and flowers prevail in these systems. The railway gardens in their majority (79%) were managed by migrants. The use of mineral fertilizers and pesticides prevalent in farms and railway gardens, while pesticide usage was absent in balcony and terrace gardens. The use of waste water among railway gardens accounted for 98% of the overall irrigation water usage; while the other production systems depend upon well water and tap water. Labor shortage is an increasing problem in UPA of MMR, while marketing channels are well organized and connected. In all cases studied, UPA systems contributed to food security and employment opportunities on limited land and helped to recycle organic waste. 124 pp. Englisch.

Nitrous oxide (N2O) is an environmentally impor…tant atmospheric trace gas and contributes to the anthropogenic greenhouse effect. In addition, it is a precursor to photochemical nitric oxide (NO) production in the stratosphere which leads to stratospheric ozone depletion. Agriculture is considered to be the main source of anthropogenic N2O, with agricultural soils representing the single largest source due to nitrogen fertilizer applications during crop production.The purpose of this study was to examine the effects of mineral N fertilizers (N form, amount, mode of application) on N2O emissions from fertilized croplands in north-west Germany. Therefore several field trials, one greenhouse pot experiment and two incubation experiments were conducted. Nitrous oxide fluxes were measured by means of the closed chamber method. The length of the experimental period varied between experiments from several weeks (42 days) up to one-year measurement campaigns. However, so far there is a lack of appropriate models that allow anticipating the mechanical stability of biofilms in particular during detachment processes.The main objective of this work was the development of a hydrogel based physico-chemical and growth independent biofilm imitate to investigate mechanical, primarily fluid dynamical stresses and their influence on growth and detachment effects of biofilms. Verification was achieved by comparison with real single culture biofilms. Single culture biofilms of Pseudomonas putida KT2440 were cultivated in a biofilm tube reactor and grown on different surfaces, e.g., tube walls, surface-modified object slides, plastic and iron nettings as well as membrane filters. The establishment of on-line analytics allowed the automatic measurement of dissolved oxygen, pH, temperature and planktonic cell growth by optical density in the cultivation broth. Image acquisition of the biofilm surface supported the observation of biofilm development in terms of growth and detachment.A hydrogel based on gellan was established as simplified artificial biofilm system, which behaves like a viscoelastic fluid. The degree of cross-linking at different gellan levels was modified by the addition of mono-and divalent ions (Na+, Mg2+) and the influence on the material constants in terms of storage (G') and loss (G'') modulus was determined. Experiments and evaluation in the predefined design space were supported by Central Composite Design (CCD), an experimental design technique.The developed gellan-based hydrogel allows mimicking the mechanical properties of a biofilm excluding biological growth effects. It can now be used to validate further characterization methods or to test slowly growing biofilms where systematic errors are often smaller than the biological variances. In this thesis I underline the importance of high resolution spectroscopy in the laboratory and in space, and the symbiotic bond…between laboratory spectroscopy and radioastronomy. My work is divided in two parts. The first part concerns high resolution spectroscopy of unstable molecules in the laboratory. The second part deals with radioastronomical observations of deuterated molecules in dense cores.The rotational spectra of CO+, 13CO+ and C18O+ in the v = 0 and 1 vibrational states have been measured in the submillimeter-wave range. Furthermore, the first THz spectra of the main isotopic species have been measured. An isotopically invariant fit has been performed, and a set of independent molecular parameters has been derived. The measured and predicted high frequency transitions of CO+ will guide new astronomical observations. Furthermore, an experimental setup to produce and probe ions in the laboratory, based on a new cryogenic discharge cell, has been developed and tested. The new experiment is presented, and some possible implementations for the future are discussed.The doubly deuterated cyclopropenylidene, c-C3D2, has been detected for the first time in space toward the starless cores TMC-1C and L1544. The chemistry that leads to the formation of c-C3D2 has been studied and is discussed in this thesis. The observed abundances c-C3D2 can be explained solely by gas-phase processes, supporting the idea that c-C3H2 is an excellent probe of gas-phase deuteration. Furthermore, l-C3HD has been tentatively detected towards TMC-1C and L1544. l-C3HD is the monodeuterated isotopologue of the linear isomer of cyclopropenylidene, propadienylidene. 142 pp. Englisch.

This thesis deals with the dy…namics of magnetic vortices as well as antivortices at picosecond and nanosecond timescales. High frequency alternating fields and spin-polarized currents are used for the manipulation of the vortices¿ magnetic states. The special case of rotational excitation is investigated by time-resolved magnetic transmission x-ray microscopy, micromagnetic simulations, and analytical calculations. For magnetic antivortices we find an asymmetric response to field and current excitation that we can explain with the negative winding number of their domains. In addition, we study the dynamics of magnetic vortices in the frequency domain by means of ferromagnetic absorption spectroscopy. We find deviations from the linear dynamics that we assumed previously. These deviations are explained in the context of a nonparabolic confining potential and a critical maximum velocity of vortex motion. Absorption spectra are found to be crucially influenced by the continuous reversal of the vortex-core polarization for high amplitudes of excitation. The insight into vortex dynamics gained in this work is crucial for a successful application of vortices for data storage devices. Magnetische Singularitäten in mikroskopisch kleinen, ferromagnetischen Dünnfilmelementen könnten in der Zukunft in magnetischen Speichermedien Verwendung finden. Des Weiteren stellen sie ein ideales Modellsystem für die Untersuchung der Magnetisierungsdynamik dar. Diese Arbeit behandelt die Dynamik magnetischer Wirbel, der sogenannten Vortizes und Antivortizes, auf Zeitskalen von wenigen Pikosekunden bis zu mehreren Nanosekunden. Hochfrequente magnetische Wechselfelder und spin-polarisierte Ströme werden genutzt um die magnetischen Zustände dieser Strukturen gezielt zu verändern. Mittels zeitaufgelöster Röntgenmikroskopie, mikromagnetischer Simulationen und analytischer Berechnungen untersuchen wir den speziellen Fall einer rotierenden Anregung. Für magnetische Antivortizes erhalten wir ein asymmetrisches Verhalten bezüglich der Feld- und Stromanregung, welche auf die besondere, negative Windungszahl der Domänenstruktur zurückzuführen ist. Zusätzlich erforschen wir die Dynamik magnetischer Vortizes im Frequenzraum mittels ferromagnetischer Absorptionsspektroskopie. Hierbei finden wir Abweichungen von den zuvor entwickelten linearen Bewegungsgleichungen. Diese Abweichungen lassen sich jedoch durch die Annahme nicht-parabolischer Einschluss-Potentiale und einer kritischen maximalen Geschwindigkeit der Vortexbewegung erklären. Es stellt sich heraus, dass die gemessenen Absorptionsspektren ab einer gewissen Anregungsamplitude grundlegend durch das kontinuierliche Schalten der Vortex-Polarisation beeinflusst werden. GaAs nanowires are characterized by a large yield strength and excepti…onal mechanical properties, making them an attractive system to study the enhancement of strain effects. At nanoscale dimensions, it is possible to achieve controllable growth of different crystal structures like zincblende or wurtzite, enabling new degrees of freedom to tailor electronic and optoelectonic properties.We show that the photoluminescence (PL) of zincblende GaAs nanowires can be red-shifted by 290 meV by axially elongating zincblende GaAs nanowires by up to 3.5%, from tension to compression. Fingerprints of symmetry breaking due to the anisotropic nature of the nanowire deformation are found in the Raman spectra, where the lifted degeneracy of the phonons is resolved, and in the PL, which undergoes a more pronounced shift in tension than in compression because of the different symmetry character (heavy or light hole) of the top valence band. In wurtzite GaAs nanowires, we demonstrate a remarkable energy shift of the PL up to 345 meV by varying the axial strain over a range of 2% in tension and compression. For the first time, we show spectroscopic evidence of a direct-to-pseudodirect bandgap transition and demonstrate that light emission can be suppressed by more than three orders of magnitude. Using the Raman scattering spectra as relative strain gauge and fitting the optical transition energies to a k¿p model, we determine all band-structure parameters of wurtzite GaAs in unstrained conditions, clarifying once and for all its band structure. Quantities like the Poisson ratio along the c-axis and the phonon deformation potentials of the GaAs and AlGaAs optical phonons have also been determined. This body of results constitutes a solid foundation for understanding strain effects on the optical and electronic properties of III-V nanowires. 222 pp. Englisch.

The simulation of hypersonic flow fields in thermochemical nonequilibrium is a challenging task, as a va…riety of flow features on various time and length scales needs to be properly resolved.With this purpose in mind, a general CFD solver framework is developed in this doctoral thesis. It combines the multiscale-based grid adaptation with the necessary physical models and numerical methods for the simulation of arbitrary reaction models in thermochemical nonequilibrium. The developed tools and methods are incorporated into the QUADFLOW solver, an integrated concept of grid generation, grid adaptation and finite-volume flow solver.The modified QUADFLOW solver is then applied to pertinent applications. The injection of various cooling gases into a supersonic boundary layer demonstrates the versatility of the QUADFLOW solver at the example of a low enthalpy configuration. The simulated high-enthalpy Edney type IV and type VII shock-shock interactions represent a complex and challenging flow configuration. A high resolution of the vortex structures in the inner flow field and of the boundary layer is achieved at the same time.Für die Auslegung und Entwicklung zukünftiger Raumtransportsysteme werden Simulationslösungen benötigt, die präzise und einfach in der Handhabung sind. Die Simulation hypersonischer Strömungen im chemischen und thermischen Nichtgleichgewicht ist eine anspruchsvolle Aufgabe, da eine Vielzahl von Strömungseffekten auf verschiedenen Zeit- und Längenskalen aufgelöst werden muss.In der vorliegenden Dissertation wird eine speziell für diese Aufgabe optimierte CFD Simulationslösung entwickelt. Hierzu wird eine multiskalen-basierte Gitteradaption mit den notwendigen physikalischen Modellen und numerischen Methoden kombiniert dieerforderlich sind, um beliebige Reaktionsmodelle im chemischen und thermischen Nichtgleichgewicht zu simulieren. Die entwickelten Modelle und Methoden werden in QUADFLOW implementiert, einer integrierten Simulationslösung bestehend aus Gittergenerierung, Gitteradaption und Finite-Volumen Strömungslöser.Die modifizierte QUADFLOW Simulationslösung wird im Anschluss zur Simulation einschlägiger Anwendungsbeispiele eingesetzt. Die Kühlgaseinspritzung verschiedener Gase in eine Überschallgrenzschicht demonstriert eindrucksvoll die Vielseitigkeit von QUADFLOW am Beispiel einer Konfiguration mit geringer Enthalpie. Die simulierten Edney Typ IV und Typ VIIStoß-Stoß Interaktionen stellen komplexe und anspruchsvolle Konfigurationen mit hoher Enthalpie dar. Verification was achieved by comparison with real single culture biofilms. Single culture biofilms of Pseudomonas putida KT2440 were cultivated in a biofilm tube reactor and grown on different surfaces, e.g., tube walls, surface-modified object slides, plastic and iron nettings as well as membrane filters. The establishment of on-line analytics allowed the automatic measurement of dissolved oxygen, pH, temperature and planktonic cell growth by optical density in the cultivation broth. Image acquisition of the biofilm surface supported the observation of biofilm development in terms of growth and detachment.A hydrogel based on gellan was established as simplified artificial biofilm system, which behaves like a viscoelastic fluid. The degree of cross-linking at different gellan levels was modified by the addition of mono-and divalent ions (Na+, Mg2+) and the influence on the material constants in terms of storage (G¿) and loss (G¿¿) modulus was determined. Experiments and evaluation in the predefined design space were supported by Central Composite Design (CCD), an experimental design technique.The developed gellan-based hydrogel allows mimicking the mechanical properties of a biofilm excluding biological growth effects. It can now be used to validate further characterization methods or to test slowly growing biofilms where systematic errors are often smaller than the biological variances. Der Membran-Befeuchter reichert die Reaktionsluft mit Wasser an, das der Abluft separiert wird. Durch die vorgestellte Methode, die Computational Fluid Dynamic (CDF)-Simulationen mit Experimenten verbindet, wird die Permeabilität der Befeuchter-Membranen für Wasser bestimmt. Die Permeabilität, dimensionslose Kennzahlen und die charakteristischen Betriebsbedingungen des Brennstoffzellenaggregats liefern die Befeuchter-Geometrie und dessen Strömungswiderstand. Anhand der Baugröße und des Strömungswiderstands werden die Investition und die Betriebskosten des Befeuchters berechnet und minimiert. Übermäßige Befeuchtung der Reaktionsluft, Lastwechselvorgänge, die zu Druckänderungen führen, und die inhomogene Gasversorgung der Brennstoffzellen im Stapel können sich in der Kondensation von Wasserdampf in den Zellen auswirken. Die zum Austrag des Kondensats erforderliche Druckdifferenz wird unter Anwendung der CFD-Methode unter Berücksichtigung der Adhäsion berechnet. Aus der Druckdifferenz werden die erforderlichen Ströme an Reaktionsgasen berechnet, die den Austrag von Kondensat und infolge dessen den stabilen Betrieb sicherstellen. Die ausgetragenen Kondensattropfen werden auf der Anodenseite aus dem Wasserstoffstrom durch den Kondensatabscheider abgeschieden, weil das Anodenabgas dem frischen Wasserstoff beigemengt wird und der Eintrag von Kondensat in die Brennstoffzellen die Reaktion beeinträchtigt. This thesis deals with the dynami…cs of magnetic vortices as well as antivortices at picosecond and nanosecond timescales. High frequency alternating fields and spin-polarized currents are used for the manipulation of the vortices' magnetic states. The special case of rotational excitation is investigated by time-resolved magnetic transmission x-ray microscopy, micromagnetic simulations, and analytical calculations. For magnetic antivortices we find an asymmetric response to field and current excitation that we can explain with the negative winding number of their domains. In addition, we study the dynamics of magnetic vortices in the frequency domain by means of ferromagnetic absorption spectroscopy. We find deviations from the linear dynamics that we assumed previously. These deviations are explained in the context of a nonparabolic confining potential and a critical maximum velocity of vortex motion. Absorption spectra are found to be crucially influenced by the continuous reversal of the vortex-core polarization for high amplitudes of excitation. The insight into vortex dynamics gained in this work is crucial for a successful application of vortices for data storage devices.Magnetische Singularitäten in mikroskopisch kleinen, ferromagnetischen Dünnfilmelementen könnten in der Zukunft in magnetischen Speichermedien Verwendung finden. Des Weiteren stellen sie ein ideales Modellsystem für die Untersuchung der Magnetisierungsdynamik dar. Diese Arbeit behandelt die Dynamik magnetischer Wirbel, der sogenannten Vortizes und Antivortizes, auf Zeitskalen von wenigen Pikosekunden bis zu mehreren Nanosekunden. Hochfrequente magnetische Wechselfelder und spin-polarisierte Ströme werden genutzt um die magnetischen Zustände dieser Strukturen gezielt zu verändern. Mittels zeitaufgelöster Röntgenmikroskopie, mikromagnetischer Simulationen und analytischer Berechnungen untersuchen wir den speziellen Fall einer rotierenden Anregung. Für magnetische Antivortizes erhalten wir ein asymmetrisches Verhalten bezüglich der Feld- und Stromanregung, welche auf die besondere, negative Windungszahl der Domänenstruktur zurückzuführen ist. Zusätzlich erforschen wir die Dynamik magnetischer Vortizes im Frequenzraum mittels ferromagnetischer Absorptionsspektroskopie. Hierbei finden wir Abweichungen von den zuvor entwickelten linearen Bewegungsgleichungen. Diese Abweichungen lassen sich jedoch durch die Annahme nicht-parabolischer Einschluss-Potentiale und einer kritischen maximalen Geschwindigkeit der Vortexbewegung erklären. It combines the multiscale-based grid adaptation with the necessary physical models and numerical methods for the simulation of arbitrary reaction models in thermochemical nonequilibrium. The developed tools and methods are incorporated into the QUADFLOW solver, an integrated concept of grid generation, grid adaptation and finite-volume flow solver.The modified QUADFLOW solver is then applied to pertinent applications. The injection of various cooling gases into a supersonic boundary layer demonstrates the versatility of the QUADFLOW solver at the example of a low enthalpy configuration. The simulated high-enthalpy Edney type IV and type VII shock-shock interactions represent a complex and challenging flow configuration. A high resolution of the vortex structures in the inner flow field and of the boundary layer is achieved at the same time.Für die Auslegung und Entwicklung zukünftiger Raumtransportsysteme werden Simulationslösungen benötigt, die präzise und einfach in der Handhabung sind. Die Simulation hypersonischer Strömungen im chemischen und thermischen Nichtgleichgewicht ist eine anspruchsvolle Aufgabe, da eine Vielzahl von Strömungseffekten auf verschiedenen Zeit- und Längenskalen aufgelöst werden muss.In der vorliegenden Dissertation wird eine speziell für diese Aufgabe optimierte CFD Simulationslösung entwickelt. GaAs nanowires are characterized by a large yield strength and exceptional… mechanical properties, making them an attractive system to study the enhancement of strain effects. At nanoscale dimensions, it is possible to achieve controllable growth of different crystal structures like zincblende or wurtzite, enabling new degrees of freedom to tailor electronic and optoelectonic properties.We show that the photoluminescence (PL) of zincblende GaAs nanowires can be red-shifted by 290 meV by axially elongating zincblende GaAs nanowires by up to 3.5%, from tension to compression. Fingerprints of symmetry breaking due to the anisotropic nature of the nanowire deformation are found in the Raman spectra, where the lifted degeneracy of the phonons is resolved, and in the PL, which undergoes a more pronounced shift in tension than in compression because of the different symmetry character (heavy or light hole) of the top valence band. In wurtzite GaAs nanowires, we demonstrate a remarkable energy shift of the PL up to 345 meV by varying the axial strain over a range of 2% in tension and compression. For the first time, we show spectroscopic evidence of a direct-to-pseudodirect bandgap transition and demonstrate that light emission can be suppressed by more than three orders of magnitude. Using the Raman scattering spectra as relative strain gauge and fitting the optical transition energies to a k¿p model, we determine all band-structure parameters of wurtzite GaAs in unstrained conditions, clarifying once and for all its band structure. Quantities like the Poisson ratio along the c-axis and the phonon deformation potentials of the GaAs and AlGaAs optical phonons have also been determined. Eine Reduktion der Versetzungsdichte und der Verspannun…g in diesem Material ist entscheidend für die Anwendung als Basisschichten für lichtemittierende Bauelemente ¿ und damit die Grundlage für effiziente UV-LEDs.Zunächst wurde glattes und versetzungsreduziertes AlN auf Saphir reproduzierbar abgeschieden. Mit einer Optimierung der Wachstumsbedingungen konnte die Schraubenversetzungsdichte in den AlN-Schichten signifikant reduziert werden. Allerdings wird dadurch eine Prozessinstabilität verursacht, die zu einer Rauigkeit führen kann. Dies lässt sich durch die Existenz von Inversionsdomänen erklären. Mit einem optimierten Desorptionsprozess konnten reproduzierbar glatte AlN/Saphir-Schichten als Grundlage für alle folgenden Untersuchungen hergestellt werden. Da die Stufenversetzungsdichte in diesen Schichten noch zu hoch ist, um effiziente LEDs zu realisieren, wurde der Ansatz des epitaktischen lateralen Überwachsens mit AlN verfolgt. Mittels dieser Methode wurde eine Versetzungsreduktion um mehr als eine Größenordnung erreicht, die mit verschiedenen Charakterisierungsmetho-den verifiziert wurde. Zudem konnten der Einfluss verschiedener Wachstumsparameter auf das Lateralwachstum von AlN bestimmt und verschiedene Motive und deren Auswirkungen auf das Koaleszenzverhalten untersucht werden. Dabei konnten verbesserte Eigenschaften von UV-LED Hetero-strukturen nachgewiesen und die Eignung der Schichten als Basis für UV-B LEDs verifiziert werden. Für die Entwicklung von UV-C LEDs konnten auf den entwickelten Templates n-leitfähige AlGaN-Schichten mit einem Al-Gehalt von über 77 % realisiert und eine Steigerung der internen Quanteneffizienz von AlGaN-Quantenfilmen erreicht werden. Diese Ergebnisse legen die Grundlage für die Realisierung von kurzwelligen UV-C LEDs mit Emissions-wellenlängen unter 250 nm. 164 pp. Deutsch.

Eine Behandlung von P. aeruginosa-Infektionen wird zunehmend erschwert und es wird immer wichtiger neue Ansatzpunkte für Vermeidungsstrategien zu entwickeln. In dieser Arbeit wurde ein systembiologischer Ansatz angewandt, um den zentralen Kohlenstoffmetabolismus von P. aeruginosa zu charakterisieren und Adaptationseigenschaften sowie die Pathogenität des Mikroorganismus umfassend zu untersuchen. Mit Hilfe der 13C-metabolischen Flussanalyse konnten zentrale in vivo-Enzymaktivitäten und Stoffwechselwege des Modellorganismus P. aeruginosa PAO1 aufgeklärt und anschließend die Analysemethode auf 11 uropathogene klinische P. aeruginosa-Isolate aus Harnwegsinfektionen sowie 12 Isolate von Patienten mit einer katheterassoziierten Harnwegsinfektion übertragen werden.
