Drug- and gene delivery to the brain is highly restricted by the vascular barriers of the brain, denoted by the blood-brain barrier (BBB) and the blood-cerebrospinal fluid (CSF) barriers. Among these barriers, BBB is the main limiting factor. It is composed by the brain capillary endothelial cells (BCECs). The BCECs barrier function is supported by astrocytes, pericytes and neurons to form the blood-brain barrier. BCECs are very tightly connected to each other by tight junctions. Apart from the essential substrates needed to nourish the brain, small and/or lipophilic molecules are free to diffuse into the brain. However most pharmacologically active drugs and gene fragments are too large to enter the brain. Various kinds of drug-carriers have been constructed for delivery of large substances to the brain. Such drug-carriers have to be able to successfully carry their cargo through the BCECs and into the brain. For testing the ability of drug-carriers to deliver their cargo into the brain, investigators have constructed different in vitro BBB models, consisting of BCECs that express the main characteristics of the BBB in vivo.In the first part of the thesis the ability of two drug-carriers, pullulanspermine and SPIOs, to mediate transfection of BCECs or transcellular transport through BCECs in vitro was studied.

This book is a contribution to the understanding of the mechanisms underlying central sensitization of spinal nociception in humans. The aims of this project were to explore different models of central sensitization in humans and to assess them objectively using nociceptive withdrawal reflexes and reflex receptive fields. The book contains four chapters. The first chapter presents the necessary background knowledge on central sensitization, the aim of the project and an overview of the dissertation.The second chapter depicts the methodology used for objective assessment of central sensitization, using nociceptive withdrawal reflexes and reflex receptive fields. The third chapter describes the human models for central sensitization studied during this project, and the thesis is completed with a fourth chapter with a brief conclusion and future perspectives.

During this project five months, between April and August 2011, were spent at Duke University, Durham, NC, USA in collaboration with Dr. Warren M. Grill. This thesis investigates the basic methodology behind the human nociceptive withdrawal reflex (NWR). The aims of this thesis were to investigate this methodology as a potential way of reducing the variability of NWR assessment and, if possible try, to discover methods which could help to reduce this variability. In order to fulfill these aims electrophysiological experiments, advanced mathematical modeling and anatomical studies were applied to investigate the elicitation and recording of the NWR. This thesis contains four chapters. The Introduction presents the reader to the background and motivation for this project and gives a general overview of the PhD thesis.The Methods chapter introduces the used methods and elaborates the background for choosing them. The Results chapter presents and discusses the main findings in this thesis; in addition, all these findings are further elaborated in the original papers. The Conclusions chapter sums up the main findings and the impact on the state of the art, and finally looks at the perspectives towards future studies. The thesis is based on three original papers that all are either published in, or submitted to, peer-reviewed journals. In addition one submitted peer- reviewed paper and several conference abstracts were based on the research conducted in this project.

The success of a neural electrode is often measured by its performance in the selective stimulation of or recording from individual fascicles within a peripheral nerve without damaging the nerve. Neural prostheses are limited by the availability of peripheral neural electrodes that can be used to record the user's intention or to provide sensory feedback through functional electrical stimulation for an extended duration. Improvement in future neural prostheses may be achieved through the availability of multichannel, bidirectional neural interfaces, such as the transverse intrafascicular multichannel electrode (TIME). The aim of this thesis was to investigate the biofunctionality, biostability, and biosafety of the TIME in an animal model that bears a close neuroanatomical resemblance to the human. To this end, four research questions were formulated: 1) Is the pig model neuro-anatomically suitable for the simulation of the actions and reactions of a neural electrode in a large human-sized peripheral nerve? This question was addressed in study 1 in which a comparative morphology study was performed on the median and ulnar nerves of Danish landrace pigs and Gottingen mini-pigs.Nerve specimens were selected at different levels for analysis. The number of fascicles in each nerve was counted, and the fascicle diameters were measured. 2) How selective is the TIME compared to other intrafascicular electrodes, such as the thin-film longitudinal intrafascicular electrode (tfLIFE)? Study 2 was performed to address this question. A comparative stimulation selectivity study was performed in Danish Landrace pigs with the TIME and tfLIFE. Electrodes were implanted in the median nerve, and sequential electric stimulation was applied to individual contacts. The compound muscle action potentials of seven forelimb muscles were recorded to quantify muscle recruitment. 3) Is the TIME biostable and biofunctional for long-term implantation? This question was addressed in study 3 in which the chronic stimulation selectivity of the TIME was evaluated on Gottingen mini-pigs. 4) Does the TIME fulfil the criteria of biocompatibility and chronic stability? Study 4 was conducted to address this question. Immunohistological analysis was conducted on nerve specimens from chronic experiment pigs that had been implanted with TIMEs in study 3.

Such power consumption issues can be mitigated through cognitive radio (CR) techniques.The use of greater bandwidths through CR and in some cases the aggregation of bandwidths, as well as the opportunistic use of spectrum of more appropriate propagation characteristics or with better interference management, can reduce necessary transmission power for devices and systems. Moreover, CR techniques may achieve greater spatial/temporal awareness of connectivity options and their inherent power consumptions, as well as the power consumptions of chosen radio access characteristics. They might be able to use this awareness to dynamically switch to connections that better manage loads and interference, or might be able to minimize the power consumption of radio frequency circuitry through switching radio access technique given a required traffic load. At a simpler level, they may be able to appropriately select nearer connectivity options minimizing path loss/shadowing hence necessary transmission power, in consideration of experienced/predicted traffic loads and channel characteristics.Such capabilities are particularly relevant considering that, at least in terms of the transmission power to from-the-socket power consumption ratio, lower power equipment such as wireless LAN access points achieves far better efficiency than higher-power equipment such as macro-cell base stations. This manifestation is not only because of favorable path loss characteristics, but also because of the better efficiencies of the lower power components in such access points. CR might also assist in making other areas of life more environmentally friendly. The application of CR to intelligent transportation, for example, facilitating the sharing of information in a timely manner and with appropriate data rates among vehicles and between vehicles and roadside infrastructure through enhancing the capabilities of systems such as IEEE 802.11p, could lead to the better management of traffic signal.
