Ensuring safety-in-use of these many chemicals is a dynamic challenge, for established ones are continually being displaced by newly developed ones more acceptable to food technologists, pharma cologists, toxicologists, and changing pest-control requirements in progressive food-producing economies. These matters are also of genuine concern to increasing numbers of governmental agencies and legislative bodies around the world, for some of these chemicals have resulted in a few mishaps from improper use. Adequate safety-in-use evaluations of any of these chemicals persisting into our food stuffs are not simple matters, and they incorporate the considered judgments of many individuals highly trained in a variety of complex biological, chemi cal, food technological, medical, pharmacological, and toxicological dis ciplines.

Cutting across the traditional boundaries that have separated the subjects in past volumes on regulation is not an easy thing to do-not because it is difficult to think of what interesting topics should replace the old ones, but because it is difficult to find authors who are willing to write about areas outside those pursued in their own laboratories. Anyone who takes on the task of reviewing a broad area of interest must weave together its various parts by picking up the threads from many different laboratories, and attempt to produce a fabric with a meaningful design. Information accu mulated during the last 10 yea…rs has shown that the function of the blood-ocular barriers may be better understood if two main barrier systems are considered to exist in the eye. The blood-aqueous barrier regulates the exchanges between the blood and the intraocu lar fluids, and the blood-retinal barrier separates the neural tissue from the blood. Recent studies have shown that the blood-retinal barrier plays a fundamental role in controlling the microenvironment of the retina. Similarly, the significance of the blood-retinal barrier in retinal disease has become increasingly clear. Fluorescein angiography has demonstrated an intricate series of relationships between altera tions of the blood-retinal barrier and diverse retinal diseases, particularly vascular retinopathies and pigment epitheliopathies. Finally, in the past few years, vitreous fluorophotometry has pro vided a new and accurate index of the alteration of the blood-retinal barrier.

While it was possible to include a large variety of spectroscopic methods, inevitably some fields had to be cut short or left out altogether. Other fields have developed so rapidly that they demand full cover in an additional volume. One of the major problems, already encountered during the prepar ation of the first volumes, was to keep track of new developments and approaches which result in spectroscopic data. We have to look far beyond the area of traditional atomic spectroscopy since methods of atomic and ion collision physics, nuclear physics, and even particle physics all make important contributions to our knowledge of the static and dynamical state of atoms and ions, and thereby greatly add to the continuing fascination of a field of research which has given us so much fundamental knowledge since the middle of the last century. Yet there are signs that a well coordinated multidisciplinary research program can conquer the epidemic and, perhaps, provide the basis for preventing future epidemics. The HIV family of viruses is now better understood, both in terms of structure and function, than any other virus. Genetically engineered peptides and nucleic acids are being tested as specific treatments or vaccines against HIV infection/disease. Most prom ising are the strides which have been made in understanding those aspects of human behavior which have contributed to the spread of HIV infection and which must be substantially modified if AIDS is to be controlled and eventually eradicated. The basis of that understanding has roots in a diverse set of disciplines which have converged in the work presented in this book.

Moreover. oral ganciclovir, cidofovir, and sorivudine are not far behind. Clinical trials with the second-site reverse transcriptase inhibitors and the protease inhibitors for HlV infection are proceeding rapidly and on a broad scale, and the preliminary results would suggest that several of these classes of drugs will be licensed as well. Despite this optimism, however, there is increasing evidence that antiviral-resistant strains of pathogenic viruses will be a significant problem, perhaps especially with therapy of HIV infection, and there remains a desperate need for improved drugs (with either improved efficacy or decreased toxicity, or both) for CMV and HIV infections. This book is the edited proceedings of the Fourth Triennial Conference on Antiviral Chemotherapy, held in San Francisco, in November 1994. The conference was sponsored by the University of California, San Francisco, and co-sponsored by the International Society for Antiviral Research (ISAR), the Macfarlane Burnet Centre for Medical Research in Melbourne, Australia, and the Australian National Centre for HIV Virology Research. The conference had been organized to present an overview of the field of antiviral chemotherapy.
