These papers, selected by the editors and reviewed by persons knowledgeable in the field, are those of the symposium type and not those pertaining to specific research topics that one would expect to be submitted to a journal. It is the opinion of the committee that this type of publication has an important place in the literature. The relatively large number of papers would result in quite a sizable volume if bound in one set of covers. For this reason, and to present the material in areas of more specific mterest, Volume 7 was divided into two parts, Part A, Physical-Inorganic, and Part B, Physical-Organic Developments. The automation of Edman chemistry (3) played a major role in the determination of polypeptide structures. Important modifications of Edman chemistry include the solid-phase approach by Laursen in 1971 (4) and the use of modified Edman reagents such as 4-N,N-dimethylaminoazobenzene-4'-isothiocy- ate (DABITC) for manual sequencing by Chang et al. (5) in 1976. A second major breakthrough in the analysis of polypeptides was automated amino acid analysis described by Spackman et al. in 1958 (6). However, during the period from 1975 to 1980, it became increasingly clear that the amount of material required for struc tural analysis was more than could be easily isolated for the vast majority of proteins. The field was criticized for its lack of sensitive techniques for the analysis of growth factors, immune modulators, membrane receptors, and peptide hormones. In addition, very little had been done to modernize and improve the original instruments introduced in the mid-1960s. The first indications of improved instrumentation for Edman chemistry came from Wittmann-Liebold's laboratory (7), followed by the introduction of a 'micro' sequencer by Hunkapiller and Hood in 1978 (8). The movement toward improved instrumentation culminated in the 'gas'-phase sequencer of Hewick et al. (9) in 1981.…

More recently attention has turned to the potential importance of the polyglutamyl derivatives of methotrexate that have been detected in normal and malignant cells both in vitro and in vivo. The biochemical trans formation of this important chemotherapeutic agent is of particular significance since polyglutamyl derivatives of methotrexate are not only potent inhibitors of the target enzyme, but have quite different cellular pharmacokinetics than the parent monoglutamate. Hence, nearly three and a half decades after 4-aminoantifolates were first applied clinically in the treatment of human neoplasms, we have begun to appreciate a new dimension in antifolate pharmaco logy which may have profound implications for our understanding of the mechanism of the cytotoxicity and selectivity of this class of agents. With the development of highly sensitive methodology for the rapid detection of folyl and antifolyl polyglutamates, it is now possible to define in depth the intracellular transformation of these agents and their role in determining antifolate action against normal and malignant cells. This information will very likely influence how regimens with methotrexate and related antifolates will be further developed and employed clinically.…

However, just as it has been recognized that primary substance abuse is frequently associated with other diagnosable psychi atric disorders, such as sociopathy or attention deficit disorder (residual type) and that the origins of substance abuse are multivariate, we have also begun to become aware that many other individuals in our society with psychiatric or other problems also suffer, to varying degrees, from substance abuse. These problems may be considered secondary by vari ous specialists or treatment personnel; but nevertheless, they are prob lems, and what disorder is primary or secondary in a given individual may often be very difficult to determine in a meaningful fashion. Thus, within the past decade, research studies have reported significant inci dences of substance abuse/or misuse in high school and college-aged populations, in medical populations, and in individuals with other psy chiatric disorders such as schizophrenia, depression, and the anxiety and personality disorders. Yet to date little has been done to bring together and systematize this widely scattered data that describes the presence of substance abuse problems in various populations.…

It is sober, concerned to be clear and deals with specific problems. It is therefore understandable that, in a philosophical climate in which Husserl's masterpiece has come to be regarded as a mere stepping stone on the way to his later Phenomeno logy, or even to the writings of a Heidegger, Reinach's contributions to exact philo sophy have been all but totally forgotten. The topics on which Reinach wrote most illuminatingly, speech acts (which he called 'social acts') and states of affairs (Sachverhalte ), as well as his realism about the external world, have come to be regarded as the preserve of other traditions of exact philosophy. Like my fellow contributors, I hope that the present volume will go some way towards correcting this unfortunate historical accident. Reinach's account of judgements and states of affairs, an account that precedes those of Russell and Wittgenstein, his 1913 treatment of speech acts, his reinter pretation of Hume and aspects of his legal philosophy are the main philosophical topics dealt with in what follows. But his analysis of deliberation as well as his work on movement and Zeno's paradoxes get only a passing mention.…

Photosynthetic reaction centers are intrinsic membrane proteins which, by performing a photoinduced transmembrane charge separation, are responsible for the conversion and storage of solar energy. Since the pioneering work of Reed and Clayton (1968) on the isolation of the reaction center from photosynthetic bacteria, optical spectroscopy with polarized light has been one of the main tools used to investigate the geometrical arrangement of the various chromophores in these systems. The recent elucidation by X-ray crystallography of the structure of several bacterial reaction centers, a breakthrough initiated by Michel and Deisenhofer, has provided us with the atomic coordinates of the pigments and some details about their interactions with neighboring aminoacid residues. This essential step has given a large impetus both to experimentalists and to theoreticians who are now attempting to relate the X-ray structural model to the optical properties of the reaction center and ultimately to its primary biological function.…
