Neuware -In this volume is presented the proceedings of a NATO Advanced Study Institute (ASI) on the theme of Electromagnetic Modelling and Measurements for Analysis and Synthesis Problems. The ASI was held at 11 Ciocco, Castelvecchio Pascoli, Tuscany, Italy, August 10th - 21st, 1987. It has been my good fortune to act as co-director of two of Jozef's previous ASIs, and so I am well acquainted with the JKS format for ASIs. As participants will realise, I did not attend this ASI, and so I only have a partial appreciation of the programme. In particular it has not been possible to include transcripts of any panel discussions which may have taken place. Readers may recall that such transcripts have formed a most interesting and useful part of previous ASI proceedings edited by Jozef Skwirzynski, and helped to convey the spirit of the meetings. Unfortunately it has proved impossible to locate the tapes, despite the best efforts of Jozef's assistant, Barry Stuart. Thus, while John Glucker's paper, 'Images of Plato in Late Antiqu ity,' raises, by means of the Platonic example, the problem of interpreta tion of ancient texts, suggesting the assignment of proper weight to the creator of the tradition and not only to his many later interpreters in assessing the proper relationship between originator and commentators, Abraham Wasserstein's 'Hunches that did not come off: Some Prob lems in Greek Science' illustrates the long-lived Whiggish tradition in the history of science and mathematics. In a second printing in 1988 it was possible to complete a few literature references and to correct some misprints. However, vigorous research has taken place over the past five years in various areas of tropical climate dynamics, especially in the atmosphere-ocean mechanisms of climate anomalies, climate prediction, ocean circulation, and paleoclimates. Promising progress has also been made in the application of general circulation modelling to tropical climate problems. In the present second edition, named 'Climate Dynamics of the Tropics', I have attempted to incorporate much of the recent work to late 1990. Chapters 8 and 9 have been essentially re-written, and major additions have been made to Chapters 4 and 12 in particular. I would like to acknowledge the continued support by the U.S. National Science Foundation over the past five years. B. Parthasarathy, Poona, and H. Lessmann, San Salvador, sent me updates of data series not easily accessible. I have benefitted from discussions with numerous colleagues in the United States and overseas. A particular advantage of a cytochemical approach to the investiga tion of mitochondrial disorders is that it allows the mosaic distribution of certain of these defects to be detected, whereas the tissue homogeniza tion involved in conventional enzyme assays or immunoblotting precludes this. A further advantage of MEA or ICC is that only small amounts of tissue are needed, which is important since many of the affected patients are infants or small children. The main aim of this communica tion is to outline ways in which these techniques can be used in the diagnosis and further investigation of mitochondrial disorders. Reference will be made not only to those situations in which MEA and ICC offer advantages over standard enzyme asays and immunoblotting but also to contexts in which the reverse applies. 4. 2 MATERIALS AND METHODS Muscle biopsies for cytochemical investigation were snap-frozen using isopentane cooled to - 150°C in liquid nitrogen. Samples were stored in heat-sealed polythene packets in the vapour phase of liquid nitrogen containers. 4. 2. 1 Microphotometric enzyme assays Frozen sections 8 Jlm thick were cut using a Reichert-J ung Frigocut cryostat microtome equipped with motor-driven cutting action to maintain maximal reproducibility of section thickness.
