However, what limits should be set upon those sacrifices, and how those limits should be determined, have long been issues of debate. In recent years this debate has intensified. There are a variety ofreasons for this; to mention only three: (1) Over the years the US health care delivery system has become increasingly complex, and with this complexity there has come a need for more and more people to have access to patients' medical records. With each transference of information, breaches in confidentiality become more likely. (2) Medical costs have risen at an alarming rate. This makes health insurance a virtual necessity for adequate medical care, and people worry that they will be denied employment and/or medical cov erage if certain sorts of medical information are not kept strictly confi dential. (3) Finally, many medical records are now kept in computer files, and the impossibility of guaranteeing confidentiality for files of this sort is a constant worry.

Research into the molecular basis of the MDs by a…c- bination of positional cloning and candidate gene analysis has provided the basis for a reclassification of these disorders, with genetic and protein data augmenting traditional clinically based nomenclature. These findings have brought insights into the molecular pathogenesis of MD, with an increasing number of potential pathways involved in arriving at a dystrophic phenotype. Some common themes can be recognized, however, including the involvement of five members of the dystrophin-associated complex (dystrophin and four sarcoglycans) in different types of MD, and the involvement of two nuclear envelope proteins in producing an Emery-Dreifuss MD phenotype. Other d- ease-associated genes appear to cause MD in a completely unrelated way, such as the involvement of calpain 3 in a form of limb-girdle muscular dystrophy. Section 1 of Muscular Dystrophy: Methods and Protocols reviews tra- tional strategies used to identify MDs. Meantime, techniques developed as a result of the research strategies described previously have become an integral part of the management of many patients with MD and their families, and these techniques are addressed in Sections 2 (DNA-based tests) and 3 (p- tein-based analyses). The continued effort to translate this enhanced und- standing into a molecular cure or treatment for MD is reviewed in Section 4.

One important aspect was that the conference did not concentrateon any particular group or type ofmaterial; rather the aim was to attract contributions from workers engaged in ageing studies with as wide a range of materials as possible. In this way, it was hoped that delegates could interactwith and learnfrom those whom they perhapswould not normally come across and that metallurgists could learn from polymer scientists, ceramicists could talk to modellers, and so on, in this important field. A read through the diverse papers contained within these proceedings will confirm that this aim was happily satisfied. For studying such equations, a new unified technology has been developed, involving analysis on infinite-dimensional manifolds. A striking applications of the new technology is Donaldson's theory of 'anti-self-dual' connections on SU(2)-bundles over four-manifolds, which applies the Yang-Mills equations from mathematical physics to shed light on the relationship between the classification of topological and smooth four-manifolds. This reverses the expected direction of application from topology to differential equations to mathematical physics. Even though the Yang-Mills equations are only mildly nonlinear, a prodigious amount of nonlinear analysis is necessary to fully understand the properties of the space of solutions. . At our present state of knowledge, understanding smooth structures on topological four-manifolds seems to require nonlinear as opposed to linear PDE's. It is therefore quite surprising that there is a set of PDE's which are even less nonlinear than the Yang-Mills equation, but can yield many of the most important results from Donaldson's theory. These are the Seiberg-Witte~ equations. These lecture notes stem from a graduate course given at the University of California in Santa Barbara during the spring quarter of 1995. The objective was to make the Seiberg-Witten approach to Donaldson theory accessible to second-year graduate students who had already taken basic courses in differential geometry and algebraic topology. 136 pp. Englisch.
