These developing countries have taken advan tage of the basic practical methods that were so well described in the first edition of this work. It is significant to note how many centers have been established throughout the world and are now studying the basic concepts and applying them to practical problems such as the detection of genetic effects caused by exposure to chemicals. In fact, there are now toxicology training centers in twelve countries. Genetic toxicology, in addition to being investigated as a science unto itself, has been taught to people in the applied fields so that these techniques may be put to use in solving other biological problems. For these reasons, it is most useful to have an update of the basic methods and their development. The bit-serial tradition at Edinburgh University (EU) stretches back some 6 years to the conception of the FIRST silicon compiler. FIRST owes much of its inspiration to Dick Lyon, then at Xerox P ARC, who proposed a 'structured-design' methodology for construction of signal processing systems from bit-serial building blocks. Based on an nMOS cell-library, FIRST automates much of Lyon's physical design process. More recently, we began to feel that FIRST should be able to exploit more modern technologies. Before this could be achieved, we were faced with a massive manual re-design task, i. e. the porting of FIRST cell-library to a new technology. As it was to avoid such tasks that FIRST was conceived in the first place, we decided to move the level of user-specification much nearer to the silicon level (while still hiding details of transistor circuit design, place and route etc. , from the user), and by so doing, enable the specification of more functionally powerful libraries in technology-free form. The results of this work are in evidence as advances in serial-data design techniques, and the SECOND silicon compiler, introduced later in this book. In 1965 it was costly and time consuming to analyze circuits consisting of a half-dozen transistors. By 1975 circuits composed of hundreds of transistors were analyzed routinely. Today, simulation capabilities easily extend to thousands of transistors. Circuit designers use simulation as routinely as they used to use a slide rule and almost as easily as they now use hand-held calculators. However, just as with the slide rule or hand-held calculator, some designers are found to use circuit simulation more effectively than others. They ask better questions, do fewer analyses, and get better answers. In general, they are more effective in using circuit simulation as a design tool. All these procedures while appearing to hold the promise of reducing the acute mortality from myocardial infarction, create the need for a planned approach to the long term management of these patients. These decisions are not always highly visible. They are made, or dinarily, within wide areas of discretion. The aims of the decisions are not always clear, and, indeed, the principal objectives of these decisions are often the subject of much debate. Usually they are not guided by explicit decision policies. Often the participants are unable to verbalize the basis for the selection of decision alternatives. Adequate information for the decisions is usually unavailable. Rarely can the decisions be demonstrated to be rational. By a rationaldecision we mean 'that decision among those possible for the decisionmaker which, in the light of the information available, maximizes the probability of the achievement of the purpose of the decisionmaker in that specific and particular case' (Wilkins, 1974a: 70; also 1969). This definition, which stems from statistical decision theory, points to three fundamental characteristics of decisions. First, it is as sumed that a choice of possible decisions (or, more precisely, of possible alternatives) is available. If only one choice is possible, there is no de cision problem, and the question of rationality does not arise. In 1975 it was first demonstrated that taurine deprivation produoed retinal degeneration in cats; more reoent studies showed that a taurine-free diet or the administration of taurine transport inhibitors caused retinal degeneration in other mammlas. At this meeting studies on three tumor viruses were discussed: the papovaviruses, the adenoviruses and the herpesviruses. The present series Developments in Molecular Virology chose to reverse this sequence by first publishing books on the herpesviruses, followed by adenoviruses, and only now the papo vaviruses. All the DNA tumor viruses gained their original reputation by serving as model systems in animal cells for studying gene expression and gene regulation, but SV40 and polyoma have been the jewel in the crown in these studies, as A phage was for the study of prokaryotes. SV40 was the first DNA tumor virus to be completely sequenced that enabled the definition of the cis controlling elements in DNA replication and transcription. I am continuously fascinated by the organization of the SV40 and polyoma genomes. Although they contain about 5000 bp that encode for only 6 to 7 proteins, the mechanisms which regulated their gene expression are varied and include almost any other type of gene regulation found today to regulate eukaryotic genes. Just to mention two: (i) the early promoter is a classical promoter that contains the TAT A, CAAT and enhancer elements, while the late promoter is devoid of these elements, and (ii) the mRNA can be structurally and functionally monocistronic or dicistronic. This hints at the versatility in the control of gene expression at the transcriptional and translational levels.
