For situations where more than one decision-maker is involved classical combinatorial optimization theory does not suffice and it is here that cooperative game theory can make an important contribution. If a group of decision-makers decide to undertake a project together in order to increase the total revenue or decrease the total costs, they face two problems. The first one is how to execute the project in an optimal way so as to increase revenue. The second one is how to divide the revenue attained among the participants. It is with this second problem that cooperative game theory can help. The solution concepts from cooperative game theory can be applied to arrive at revenue allocation schemes.In this book the type of problems described above are examined. Although the choice of topics is application-driven, it also discusses theoretical questions that arise from the situations that are studied.For all the games described attention will be paid to the appropriateness of several game-theoretic solution concepts in the particular contexts that are considered. The studies range from classifications and taxonomies of behaviors, to development of architectures which cause such group activities as flocking or swarming, and from emphasis on the role of intelligent agents in such groups to studies of learning and obstacle avoidance.There used to be a time when many robotics researchers would question those who were interested in working with teams of robots: `Why are you worried about robotic teams when it's hard enough to just get one to work '. This issue responds to that question. Robot Colonies provides a new approach to task problem-solving that is similar in many ways to distributed computing. Multiagent robotic teams offer the possibility of spatially distributed parallel and concurrent perception and action. A paradigm shift results when using multiple robots, providing a different perspective on how to carry out complex tasks. New issues such as interagent communications, spatial task distribution, heterogeneous or homogeneous societies, and interference management are now central to achieving coordinated and productive activity within a colony. Fortunately mobile robot hardware has evolved sufficiently in terms of both cost and robustness to enable these issues to be studied on actual robots and not merely in simulation.Robot Colonies presents a sampling of the research in this field. However, such an event did take place June 22-26, 1995, at an international symposium held at Stanford University to celebrate Professor Thomas Kailath's sixtieth birthday and to honor the notable con tributions made by him and his students and associates. Over the five days of the meeting, there were about 200 at tendees, from eighteen countries, more than twenty government and industrial organizations, and various engineering, mathematics and statistics faculties at nearly 50 different academic institutions. They came not only to celebrate but also to learn and to ponder the threads and the connections that Professor Kailath has discovered and woven among so many apparently disparate areas. This chapter provides an introductory overview of the rapidly evolving field of non-viral approaches for gene delivery to rnarnrnalian cells. Although currently there are fewer ongoing clinical trials using non-viral approaches than those using viral based systems, the number of non-viral trials is increasing. The long range goal of some research groups is the development of a genetically engineered artificial virus targeted to specific cells in the human body. An arurual conference, organized by Cambridge Healthtech Institute entitled 'Artificial Self-Assembling Systems for Gene Transfer', brings together researchers interested in this field [1]. Assembly of an artificial virus is very complex; other research groups aim to develop simpler delivery systems consisting of a plasmid combined with delivery agents. Viral-based systems are very successful for gene delivery, but despite their successes, viral-based systems have some geneml limitations and system-specific limitations. When employing a viml-based system, the following limitations should be considered: ¿ size limitation of the inserted gene due to packaging constraints (e. g. adenovirus, retrovirus) . ¿ potential tumorigenesis (e. g. retrovirus) ¿ potential for insertional mutagenesis (greater than plasmid based systems) ¿ potential imrnunogenicity (e. g. 540 pp. Englisch.

Some of the applications that stand to impact this field most significantly are based on recent advanc…es in the biological sciences, as demonstrated by the chapters on gene therapy, on the detection of minimal residual disease using molecular techniques, and on the use of radioimmunoconjugates targeting lymphoma and leukemia-associated antigens. Others are the results of clinical observations - e.g., the association between graft-versus-host- disease (GVHD) and durable remissions that have led to creative clinical experiments such as donor leukocyte infusions (DLI). Attempts to unravel the biological events that underlie the responses seen in patients with relapsed chronic myelogenous leukemia treated with DLI are likely to provide the basis for future refinements in this clinical approach. Hopefully, improved response rates and reduced toxicity will result. The power of the immunologic response in controlling malignant disease is underscored in the chapter on post-transplant immunotherapy. The complex immunologic process that results in clinical GVHD may be dissected and engineered to provide clinical benefits that include, in addition to its antineoplastic effects, the amelioration of its clinical manifestations. Better control of GVHD with less global immunosuppression will facilitate the use of mismatched and unrelated donors. This area of investigation perfectly illustrates the continued interplay between the laboratory and the clinic. The continued cross-fertilization of ideas between immunologists, molecular biologists and clinical investigators is likely to yield important advances in this field for years to come.Possible applications of stem cell transplantation continue to grow with the identification of alternative sources of stem cells and the potential to engineer and/orexpand the graft. Although the use of unrelated and mismatched donors continues to increase, the possibilities associated with umbilical cord blood transplantation are legion, especially if stem cells can be expanded ex vivo to provide grafts for full-sized adults. Using techniques in which contaminating malignant cells may be eliminated from autografts through positive selection, autologous transplantation may prove highly effective, especially when coupled with post-transplant immunotherapy. Some of these same methodologies have helped facilitate the use of autologous grafts for transplantation in patients with chronic myelogenous leukemia without allogeneic donors.Advances in the supportive care of transplant patients, including the pretransplant identification of those at risk from pulmonary complications and the use of cytokines to speed engraftment, have reduced morbidity and mortality to such a degree that it is appropriate to consider high-dose therapy and stem cell reconstitution in patients with nonmalignant diseases. The impressive advances that have occurred in transplantation for thalassemia are described by pioneers in their area of investigation. The burgeoning field of transplantation for autoimmune disorders, including its immunobiologic basis and soon-to- be-realized clinical potential, is also summarized. Continued progress in the use of high-dose therapy with stem cell rescue for the treatment of pediatric tumors, which derives in part from improved supportive care, is detailed.The sobering voice of the health care economists underscores the necessary limitations to our seemingly unbridled imagination. Cost- consciousness and financial know-how will need to be reflected in future study designs. Given the seemingly endless applications of our technology, strategies to insure its cost-effectiveness will be necessary.
