Different concentration and combinations of growth regulators were used in MS medium to observe the callus induction ability from anther and filament, used as explants. Among the genotypes used BARI Sarisha-7, showed the highest percentage (81.25% and 60.42%) of callus followed by Tori-7 (58.33% and 39.58%) and Agrani (41.67% and 31.25%) from filament and anther as explants. Among the treatments, highest percentage of shoot regeneration (93.75 % and 75.00%) from filament and anther were observed in MS + 4mgL-1 BAP + 1.0mgL-1 NAA (T3). BARI Sarisha-7 and Tori-7 showed better result (79.17 % & 62.50 % and 72.92% & 56.25% from filament and anther, respectively) followed by Agrani (64.58% and 47.92%) and Daulat (58.33% and 41.67%) in shoot regeneration. Highest percentage of root induction (87.50% and 75%) from filament and anther were observed in half MS + 1.0 mgL-1 IBA + 0.5mgL-1 NAA (T1). The plantlets with sufficient roots thus obtained were transferred successfully to plastic pots and subsequently to the field. 156 pp. Englisch.

Accession wise significant variation was observed among the evaluated morpho-physiological parameters. Whereas among mineral compositions K content was the highest followed by N, Ca, Mg, Na,P,Fe and Mn respectively. ISSR and EST-SSR marker based molecular characterization has truly confirmed the genetic variability among the collected P. oleracea germplasm collections and revealed 11% and 28% variation among populations with 89% and 72% variation within populations. More or less all the salinity screened 13 purslane accessions were morpho-physiologically affected by the highest salinity levels but very interestingly measured minerals and antioxidant properties were increased at the lower to moderate (8 and 16 dS m-1) salinity and tended to decrease again at the highest (24 and 30 dS m-1) salinity compared to control. It was also found that ornamental purslane contains more antioxidant properties and more tolerant to salinity over common purslane containing more minerals as well. 236 pp. Englisch.
