We aimed to isolate cDNAs ind…uced by switching Aspergillus nidulans from glucose to selected polysaccharides. We devised a Negative Subtraction Hybridization method which involved screening a library made from cDNAs from cells grown under a selected physiological condition with labeled cDNA probes from another physiological condition. In a screen for transcripts induced by switching the carbon source, 3,532 negatives were isolated. Single-channel microarray experiments were done to confirm that the majority of the negatives were indeed induced differentially. To gain insight into the regulation, we used microarrays to identify gene expression changes following a shift from glucose to pectin or glucose starvation. The findings from this study portray the useful utilization of the powerful microarray technology to study gene expression profiling under various physiological conditions. 68 pp. Englisch.

Nous avons cherché à isoler les ADNc induits en faisant passer Aspergillus nidulans du glucose à des polysaccharides sélectionnés. Nous avons conçu une méthode d'hybridation par soustraction négative qui consiste à cribler une bibliothèque constituée d'ADNc provenant de cellules cultivées dans une condition physiologique donnée avec des sondes d'ADNc marquées provenant d'une autre condition physiologique. Lors d'un criblage des transcrits induits par le changement de source de carbone, 3 532 négatifs ont été isolés. Des expériences de microréseau à canal unique ont été réalisées pour confirmer que la majorité des négatifs étaient effectivement induits de manière différentielle. Pour mieux comprendre la régulation, nous avons utilisé des microréseaux pour identifier les changements d'expression des gènes après le passage du glucose à la pectine ou la privation de glucose. Unser Z…iel war es, cDNAs zu isolieren, die durch die Umstellung von Aspergillus nidulans von Glucose auf ausgewählte Polysaccharide induziert werden. Wir entwickelten eine negative Subtraktionshybridisierungsmethode, bei der eine Bibliothek aus cDNAs von Zellen, die unter einer ausgewählten physiologischen Bedingung gewachsen sind, mit markierten cDNA-Sonden aus einer anderen physiologischen Bedingung gescreent wurde. In einem Screening nach Transkripten, die durch die Umstellung der Kohlenstoffquelle induziert wurden, wurden 3.532 Negative isoliert. Einkanalige Microarray-Experimente wurden durchgeführt, um zu bestätigen, dass die Mehrzahl der Negative tatsächlich differenziell induziert wurden. Um einen Einblick in die Regulation zu erhalten, haben wir Microarrays verwendet, um die Veränderungen der Genexpression nach einem Wechsel von Glukose zu Pektin oder nach Glukose-Starvation zu identifizieren. Die Ergebnisse dieser Studie zeigen die nützliche Nutzung der leistungsstarken Microarray-Technologie zur Untersuchung von Genexpressionsprofilen unter verschiedenen physiologischen Bedingungen. 60 pp. Deutsch.

O nosso objectivo era iso…lar os cDNAs induzidos pela mudança de Aspergillus nidulans da glucose para polissacáridos seleccionados. Concebemos um método de Hibridação de Subtracção Negativa que envolveu o rastreio de uma biblioteca feita de cDNAs a partir de células cultivadas sob uma condição fisiológica seleccionada com sondas de cDNA rotuladas a partir de outra condição fisiológica. Num ecrã para transcrições induzidas pela mudança da fonte de carbono, foram isolados 3.532 negativos. Foram feitas experiências de microarranjo de canal único para confirmar que a maioria dos negativos foram de facto induzidos de forma diferente. Para obter informações sobre a regulação, utilizámos microarrays para identificar alterações de expressão genética após uma mudança de glucose para pectina ou inanição de glucose.
