Dorpat 1849-54 (De medullae spinalis textura in ranis ratione imprimis habita indolis substantiae cinereae) von seinem ehemaligen Lehrer Heinrich Bidder 1855 an die Anatomische Anstalt in Dorpat berufen und bildete sich 1856 als Stipendiat in Deutschland weiter. 1858 kehrte er als Prosektor und a.o. Prof. an die Univ. Dorpat zurück. 1866 habilitierte er sich für Histologie an der Univ. Kiel. Seit 1867 o. Prof. der Histologie und Anatomie, 1872-74 Rektor in Kiel, lehrte er 1876-80 Anatomie, vergleichende Anatomie und Entwicklungsgeschichte in Königsberg und war 1880-1901 Ordinarius für Anatomie und 1896/97 Rektor der Univ. München. K., seit 1879 Mitglied der Deutschen Akademie der Naturforscher Leopoldina, war einer der Begründer der vergleichenden Entwicklungsgeschichte. Nach ihm wurden die Kupffer-Blase der Knochenfische und die Kupffer'sche Sichel der Vogelembryonen benannt. K. wies u.a. Gallenkapillaren in Leberstücken nach, die er mit dem später nach ihm Kupffer Reagens benannten Färbemittel darstellte. (Quelle DBE).
