The title of Delbrúck's Nobel Prize lecture refers to his paper published in 1949 (hence "twenty years" earlier) entitled "A Physicist Looks at Biology". Appendix 1 in the Nobel lecture is Delbruck's memorandum written in 1937, "Preliminary write-up on the topic 'Riddle of Life' (Berlin, August 1937)." Quoting Delbrück in his Nobel lecture: "This memorandum would appear to be a summary of discussions at a little meeting in Copenhagen, arranged by Niels Bohr, to which Timofeef-Ressovsky, H. J. Muller, and I had traveled from Berlin. These discussions occurred very much under the impact of the findings of W. M. Stanley reporting the crystallization of tobacco mosaic virus". The first printing is printed on one side of the paper only, and the lectures are separately paginated, whereas in the second printing they are printed on both sides of the paper and are continuously paginated. The first printing is spiral bound with a blue paper wrapper that is heavier than the paper of the text, whereas in the second printing it is stapled and the wrappers are of the same paper as the text leaves. Delbrück "complained frequently that his teaching duties left too little time for research. He needed time to prepare a series of lectures he was to give at Vanderbilt's School of Medicine in April and May 1944 entitled 'Problems of Modern Biology in Relation to Atomic Physics.' These presentations were reproduced without change in February 1946 and distributed to the grantees of the American Cancer Society; they remained unpublished otherwise. After describing his view of the limitations of applying atomic physics to biology, Max proceeded in these lectures to try to determine how far atomic physics could help in understanding living cells. He discussed metabolism; photosynthesis; genes, their actions and mutations; and bacterial viruses. After first dealing with what he called ordinary biochemistry ('what the cell does with substrates introduced from without'), Max introduced genetics by reviewing the steps that lead to the picture of the gene as a material particle. He described the importance of Mendel's experiments, which showed there are factors that segregate and that can be recovered unchanged from a variety of phenotypes. He explained that cytologists were able to localize these factors in sequence along chromosomes. Then he speculated about the outlook of bacterial genetics: 'We owe our present knowledge, as it were, to two lucky accidents, the bisexuality of the higher organisms and the occurrence of crossing over at meiosis. Since our knowledge of the gene is so intimately tied to those two features, we may wonder whether genes exist also in organisms where there is no sex and no crossing over, as in bacteria.' Max pointed out that genetics now required a biochemical approach: geneticists had produced 'three magnificent problems' for biochemists: 1. What do genes consist of? 2. How do they reproduce? 3. How do they act? Max thus publicly stated his appreciation of biochemistry about a month before he learned of Avery's transforming principle" (Fischer, Ernst Peter & Carol Lipson: Thinking about Science. Max Delbrück and the Origins of Molecular Biology, p. 155). These lectures should not be confused with Delbrück's well-known lecture published in 1949, "A Physicist Looks at Biology." "Über die Natur der Genmutation und der Genstruktur" is often referred to as the "Dreimänner" ["Three-Man"] paper because of its three authors, or the "green paper" because of the color of the wrappers. It is divided into four sections. "The first section, by Timoféeff-Ressovsky, describes the mutagenic effects of X-rays and gamma rays on the fruitfly Drosophila melanogaster. . . . The second section . . . is by Zimmer and applies the target theory to Timoféeff-Ressovsky's results. . . . The third section is by Max Delbrück. . . . His paper shows the maturity, judgement and breadth of knowledge of someone who had been in the field for years. It is imaginative, and sober, and its carefully worded predictions have stood the test of time" (Perutz, "Physics and the Riddle of Life," Nature, Vol. 326, 1987, pp. 556-57). The three authors of the paper "concluded that a mutation is a molecular rearrangement within a particular molecule, and the gene a union of atoms within which a mutation, in the sense of a molecular rearrangement or dissociation of bonds, can occur. The actual calculations of the size of the gene, deduced from calculations on the assumption of a spherical target, were not cogent, as Delbrück wryly admitted in his Nobel Prize lecture, but the entire approach to the problem of mutation and the gene adopted by the three collaborators was highly stimulating to other investigations" (D.S.B. 18: 922). Chapters IV and V of Erwin Schrödinger's book What is Life? (1944) are entitled "The Quantum-Mechanical Evidence" and "Delbrück's Model Discussed and Tested", and "are largely paraphrased versions of the paper by Timoféeff, Zimmer and Delbrück. . . . In retrospect, the chief merit of What is Life? is its popularization of the Timoféeff, Zimmer and Delbrück paper that would otherwise have remained unknown outside the circles of geneticists and radiation biologists" (Perutz, ibid., pp. 556, 558). An entire book has been published about this "Three-Man Paper", with an English translation: Creating a Physical Biology. The Three-Man Paper and Early Molecular Biology, edited by Phillip R.
