Die großformatigen, vielfach farbigen Tafeln zeigen die menschliche Anatomie schichtweise und mit besonderem Gewicht auf den räumlichen Beziehungen von Organen, Gefäßen, Nerven und Muskulatur; die Darstellung ist auf die praktische Anwendung in Medizin und Chirurgie ausgerichtet. Das Werk ist zugleich historisch belastet: Pernkopf war überzeugter Nationalsozialist, und für die während der NS-Zeit entstandenen Teile des Atlas wurden auch Leichen von Opfern der NS-Justiz verwendet.
