Oft ist es schwierig, insbesondere für den mehr angewandt-histochemisch tätigen Wissenschaftler und für die technischen Mitarbeiter im Labor, den Wert einer enzymhistochemischen Reaktion zu beurteilen und eine richtige Auswahl unter vorhandenen Methoden zu treffen. Hierbei soll das vorliegende Buch helfen. Es enthält nur die wichtigsten histochemischen Enzymnachweise, die alle selbst erprobt, häufig modifiziert und vielfach mit gutem Erfolg bei den verschiedensten Forschungs-und Routineuntersuchungen an normalen und pathologisch veränderten Organen eingesetzt wurden. Verfahren, mit denen keine Erfahrungen bestehen, oder die unbefriedigende Resultate erbringen, werden erwähnt, aber nicht als Methode empfohlen. Bei der Sammlung handelt es sich daher um eine speziell für die praktische Laborarbeit gedachte, kritische Auswahl enzymhistochemischer Methoden. Theoretische Gesichtspunkte finden insoweit Berücksichtigung, als sie zum Verständnis praktisch-histochemischer Probleme und wichtiger funktionell-biochemischer Vorgänge und Zusammenhänge beitragen. Hervorgegangen ist dieses Buch aus zwei unabhängig voneinander ent standenen Rezeptsammlungen, die vielfach verändert, weitergeführt und den Fortschritten der Forschung angepaßt wurden. Beide Methodensammlungen wurden in histochemischen Kursen erprobt, die wir an verschiedenen Orten und für sehr unterschiedliche Teilnehmerkreise durchgeführt haben. In diese…r Zeit verlangert sich der Darm, Zotten entstehen und das mehrreihige Epithel wird hochprismatisch einschichtig (Hummel, 1935; Patzelt, 1936; Kammeraad, 1942; Behnke, 1963a, b; O'Connor, 1966; Deren, 1968; Voll rath, 1969; Shervey, 1973). 1m Elektronenmikroskop erkennt man, daB die Enterocyten ihr terminales Netzwerk und Mikrovilli ausbilden; ferner nehmen Lysosomen, Mitochondrien, endoplasmatisches Reticulum und Golgi-Apparat zu (Behnke, 1963a, b; Dunn, 1967; Hayward, 1967a, b; Vollrath, 1969; Williams und Beck, 1969a). Histochemisch und biochemisch lassen sich dann in Lysosomen und im Biirstensaum des Darmepithels erstmalig Glykosidasen, Phosphatasen und Peptidhydrolasen nachweisen, deren Aktivitat gemeinsam mit der der schon vorher vorhandenen cytoplasmatischen und mitochondrialen Oxydoreductasen in den letzten Tagen der Pranatalzeit auffallig ansteigt (Cohen, 1957; Semenza, 1968; Vollrath, 1969; Williams und Beck, 1969a; Gossrau, 1973b, g; Shervey, 1973). Die weitgehende Differenzierung versetzt den Diinndarm bereits vor der Geburt in die Lage, u. a. mit dem Fruchtwasser intrauterin angebotene Fette und Proteine zu resorbieren (Vollrath, 1969; Williams und Beck, 1969a; Orlic und Lev, 1973). Postnatal entwickelt sich das Darmepithel weiter, wahrend gleichzeitig Be standteile der Muttermilch resorbiert werden. Morphologisch treten im proxi malen Intestinum mit Fett gefiillte Enterocyten und distal Saumzellen mit proteinhaltigen Riesenvacuolen auf (v. Mollendorff, 1925; Clark, 1959).
