These were ketolactase test, growth on glucose peptone agar, growth on Hofer's alkaline broth, growth under anaerobic conditions, hydrogen sulphide production, tolerance to temperature extremes and heat resistance, all of which were negative. Hence it was confirmed as Rhizobium. Effect of AM fungi and Rhizobium was studied using Leonard jar experiment and open pot cultures for both species of Phaseolus. Tissue culture studies were done on MS (1962) and BLD (1966) medium supplemented with growth regulators. The amount of proteins, polyphenols, nucleic acids and peroxidases were estimated from dual cultures and calli alone. 80 pp. Englisch.

La caracterisation des deux isolats de rhizobiums a ete rea…lisee a partir de cultures agees de 24 a 48 heures. Des tests biochimiques ont ete effectues. Il s'agissait du test de la cetolactase, de la croissance sur gelose au glucose-peptone, de la croissance sur bouillon alcalin de Hofer, de la croissance dans des conditions anaerobies, de la production d'hydrogene sulfure, de la tolerance aux temperatures extremes et de la resistance a la chaleur, qui se sont tous reveles negatifs. Il a donc ete confirme qu'il s'agissait de Rhizobium. L'effet des champignons AM et du Rhizobium a ete etudie a l'aide d'une experience en bocal Leonard et de cultures en pot ouvert pour les deux especes de Phaseolus. Des etudes de culture tissulaire ont ete realisees sur des milieux MS (1962) et BLD (1966) enrichis en regulateurs de croissance. Rhizobien wurden aus beiden Phaseolus-Arten auf Hefeextrakt-Mannitol-Agar (YEMA) mit Kongorot isoliert. Die Charakterisierung beider Rhizobienisolate erfolgte anhand von 24 bis 48 Stunden alten Kulturen.…Es wurden biochemische Tests durchgefuehrt. Dabei handelte es sich um den Ketolactase-Test, das Wachstum auf Glucose-Pepton-Agar, das Wachstum auf Hofer-Alkalibruehe, das Wachstum unter anaeroben Bedingungen, die Schwefelwasserstoffproduktion, die Toleranz gegenueber extremen Temperaturen und die Hitzebestaendigkeit, die alle negativ ausfielen. Somit wurde die Zugehoerigkeit zu Rhizobium bestaetigt. Die Wirkung von AM-Pilzen und Rhizobium wurde anhand eines Leonard-Glas-Experiments und offener Topfkulturen fuer beide Phaseolus-Arten untersucht. Gewebekulturstudien wurden auf MS- (1962) und BLD- (1966) Medium durchgefuehrt, das mit Wachstumsregulatoren ergaenzt wurde. Glomus was dominant with 31 species. It was followed by 8 species of Scutellospora, 6 species of Acaulospora, 2 spe…cies of Gigaspora and 1 species of Entrophospora. Glomus aggregatum, G. callosum, G. delhiense, G. fasciculatum, G. intraradix, G. mosseae, Scutellospora calospora and S. fulgida were most common. Maximum number of AM propagules /100 g of rhizosphere soil were 837 and 394 in non rhizosphere soil. Cent percent root infection was recorded on 150th day. Number and surface area of leaves, GVI and biomass was more in experimental than control plants during salt and water stress. Chlorophyll and total soluble carbohydrates were more in experimental than control plants during salt and water stress. DNA and RNA, protein, proline and reducing sugars content was more in control than experimental plants during salt and water stress. The 6 Gingerol was more in experimental than control plants. Plants treated with mycorrhizae protected plants from water and salt stress up to certain limit as compared to control plants.
