La mayoría de las enfermedade…s oculares se tratan con la aplicación tópica de gotas para los ojos. Después de la instilación de una gota para los ojos, típicamente menos del 5% del fármaco aplicado penetra en la córnea y llega a los tejidos intraoculares, mientras que una fracción importante de la dosis instilada se absorbe y entra en la circulación sistémica. La baja capacidad del área precorneal da como resultado un volumen de gota óptimo de aproximadamente 20 µl y el riesgo de efectos sistémicos adversos de los fármacos debido a la absorción sistémica a través de la mucosa nasal. El presente trabajo se realiza reduciendo la punta del gotero insertando un capilar de vidrio. Most ocular diseases are treated with topical application of eyedrops. After instillation…of an eyedrop, typically less than 5% of the applied drug penetrates the cornea and reaches intraocular tissues, while a major fraction of the instilled dose is absorbed and enters the systemic circulation. The low capacity of the precorneal area resulting in an optimal drop volume of about 20 µL and the risk of adverse systemic effects of drugs due to systemic absorption via the nasal mucosa. The present work is carried by reducing the dropper tip by inserting glass capillary. It has a short half life and low oral bioavailability of 5%. Therefore, the purpose of this research was to develop unidirectional Bucco- adhesive films of Buspirone Hydr…ochloride by solvent casting technique using full factorial designs. HPMC K15M and Eudragit RL-100 were used as polymers in different proportion. Polyethylene glycol 400 was used as plasticizer and Sodium lauryl sulphate was used as permeation enhancer in different concentration. The physicochemical compatibility of the drug and the polymers was studied by FT-IR spectroscopy. The results suggested no physicochemical incompatibility between the drug and the polymers. In full factorial designs, total amount of polymer (X1) and percentage of HPMC K15M (X2) was kept as a independent variables. After wards, statistically optimization process was carried out and two optimized formulation (OF1 and OF2) was developed. The results of optimization formulation showed greater degree of % similarity with predicted values. Stability studies of two optimized formulations were carried out at room temperature as per ICH Q1C guidelines. 148 pp. Englisch.

Die meisten Augenkrankheiten werden mit topischer A…nwendung von Augentropfen behandelt. Nach der Instillation eines Augentropfens dringen typischerweise weniger als 5% des applizierten Arzneimittels in die Hornhaut ein und erreichen das intraokulare Gewebe, während ein Großteil der instillierten Dosis absorbiert wird und in den systemischen Kreislauf gelangt. Die geringe Kapazität des präkornealen Bereichs führt zu einem optimalen Tropfenvolumen von etwa 20 µl und dem Risiko nachteiliger systemischer Wirkungen von Arzneimitteln aufgrund systemischer Absorption über die Nasenschleimhaut. Die vorliegende Arbeit wird durchgeführt, indem die Tropfenspitze durch Einsetzen einer Glaskapillare reduziert wird.
