The asymmetric screening matrix, a D-optimal design, was used for finding out the significant formulation factors. Tablets were obtained by wet granulation of drug with HPMC K100M and various excipients, followed by compression and were evaluated for buoyancy lag time,Q 6h,Q 24h and floating duration, during screening study. The graphical analysis of coefficients was done to find out significant factors. The response surface modeling (RSM) was performed with significant factors obtained from screening study.The D-optimal design was used for RSM and various prediction plots and contour plots were used for RSM. The desirability function was used to combine all the responses for optimization. The Nelder-Mead simplex method was applied for optimization of formulation. The optimized formulation was evaluated for diff. parameters and compared with marketed formulation.Various model dependent and model independent drug release parameters of optimized formulation were comparable to that of marketed formulation. 132 pp. Englisch.
