Er studierte an der University of North Carolina at Chapel Hill, war Schauspieler bei den Carolina Playmakers und studierte dann weiter an der Harvard University. 1924-29 war er dort Dozent für amerikanische Literatur, was er jedoch aufgab, um sich ganz dem Schreiben zu widmen. Sinclair Lewis würdigte ihn in seiner Nobelpreisrede 1930 mit den Worten: Da ist Thomas Wolfe, ein Junge von, ich glaube, dreißig Jahren, dessen einziger Roman Look Homeward, Angel! an die Seite unserer besten Literaturwerke gestellt werden kann, eine kolossalische Schöpfung von tiefer Lebenslust. Thomas Wolfes Haus in Asheville, North Carolina "Wolfe unternahm sechs Reisen nach Europa und war während der Olympischen Sommerspiele 1936 in Berlin, wo er seinen deutschen Verleger Ernst Rowohlt traf.
