"A young Danish scientist . . . has bequeathed to posterity a most attractive and important thesis in the form of a monograph which, even for the sake of its illustrations alone, deserves more notice than it usually receives. This is Mikroskopsike Undersögelser af Nervesystemet, and the author was A. H. Hannover. The book was published in Copenhagen in 1843 and translated into French the following year [offered here]. It was in 1838 that the Royal Society of Sciences of Denmark, in view of the invention of the achromatic microscope, had offered a prize for the best answer to the question: 'What results can physiology deduce from recent microscopic researches on the nervous system?' So when Hannover went in 1839 to study anatomy and physiology in Berlin, Müller gave him 'the use of the university microscope' and encouraged him to find the answer. After a year in Berlin, Hannover returned to Copenhagen where he used a microscope with the then good magnification of 340. Hannover, moreover, introduced a new method which was important. This was to use as a fixative a solution of chromic acid, of such a strength as to be pale straw in colour, as Kölliker later described it. . . . As a result, Hannover's preparations of nerve cells show processes extending away for a hitherto unprecedented distance, all recognizable at once by the modern eye for what they are. He dislike the term ruban primitif for the axis cylinder, because, as he could see by his new methods, it is not a ribbon, but a free-floating entity inside the sheath" (Liddell, "Discovery of Reflexes", pp. 11-12). "During his early studies in the 1830's, Hannover began using chromic acid to fix and harden tissue. This substance not only preserved the tissue against water-induced distortions but also made it easier to cut thin sections for viewing under the microscope. In his thesis (published in Danish in 1843 and in French in 1844, he clearly illustrated nerve cells giving rise to a single large process ('nerve fiber'), thus supporting Remak's suggestion. He also differentiated between the nerve fiber (i. e., axon) and other processes (i. e. dendrites)" (Shepherd, "Foundations of the Neuron Doctrine", p. 26).
