Dans l une d elles, au sud-ouest de la ville, la peinture murale a été privilégiée comme en témoigne le célèbre tombeau des Trois Frères. Daté des iie-iiie s. ap. J.-C., il a été découvert à la fin du xixe siècle. Son plan en T renversé à trois exèdres caractérise ces tombeaux hypogées qui recueillaient des centaines de corps. Une inscription gravée au-dessus de la porte d entrée nomme les trois frères fondateurs, Nama in, Malê et Saedi. Leur famille s était réservé l exèdre du fond ornée d un décor peint couvrant la voûte et les parois. L iconographie est ici en grande partie d inspiration gréco-romaine. Sur les trois parois principales des Victoires ailées posées sur des globes soulèvent des médaillons où figurent les bustes des défunts, des hommes sobrement vêtus, des femmes arborant hautes coiffes et bijoux. Sur la lunette, une vaste composition sur fond vert représente Achille à Skyros : Ulysse et ses compagnons viennent de le démasquer sous son déguisement féminin et la vraie nature du héros se révèle. Au centre de la voûte, un médaillon figure l aigle enlevant Ganymède, le jeune berger phrygien aimé de Zeus. L arc d entrée porte un réseau de cercles sécants, et les piédroits de grandes figures féminines, l une portant son enfant, toutes deux accompagnées d inscriptions. Entre 2004 et 2009, des campagnes d étude et de documentation ont été entreprises par la mission syro-française à la demande de la Direction Générale des Antiquités et des Musées. Par chance, le tombeau n avait jusque-là fait l objet que de consolidations de structures, sans modifier l état des peintures car leur grande fragilité les avait préservées de nettoyages et de repeints. L état des lieux a été conduit de manière pluridisciplinaire : topographie, magnétométrie, épigraphie araméenne, constat d état, analyses des pigments et du support, documentation photographique et relevé graphique des décors. En mai 2015, la prise de Palmyre par Daech et l occupation du tombeau comme bureau habité l ont altéré. Les dégâts constatés dès mars 2016 touchent sa structure, plus gravement son décor peint sensible à l humidité, les parties figurées ayant été badigeonnées.
