Die Vitrifikation intrazellulärer Moleküle kann die durch Kristallisation verursachten Probleme der Kryokonservierung überwinden. Um eine Vitrifizierung zu erreichen, muss eine enorme Wärmeabfuhr von etwa 10^6 C/s auf die Zellen angewendet werden, was derzeit für biologische Materialien nicht möglich ist. In dieser Forschungsarbeit untersuchten wir eine neue Methode mittels mikrofluidischer Geräte, die von Dr. Mehmet Toner vorgeschlagen wurde, um Zellen mit CPAs vorzukonzentrieren und dabei die Zellen einer wesentlich geringeren mechanischen und osmotischen Belastung auszusetzen als bei herkömmlichen Methoden. Die neue Methode basiert darauf, Zellen in wässrigen Tröpfchen einzufangen und die CPA-Konzentration im Tröpfchen durch Anpassung der Temperatur des Systems zu steuern. Durch Finite-Elemente-Analysen des mikrofluidischen Systems haben wir festgestellt, dass es mit der neuen BioMEMS-basierten Methode möglich ist, Säugetierzellen mit CPA in 3 Minuten auf das Zehnfache der ursprünglichen CPA-Konzentration vorzukonzentrieren.…

New method is based on trapping cells into aqueous droplets and controlling the CPA concentration in the droplet by adjusting temperature of the system.
