Ihre Gesamtzahl beträgt ca. 3.500. Sie kommen vor allem aus dem Gebiet der antiken Stadt Kos, dem weltberühmten Heiligtum des Asklepios vor den Toren der Stadt und aus den Demen Halasarna, Antimachia und Isthmos. Die Arbeiten an einem Corpus der Inschriften von Kos reichen bis in das 19. Jh. zurück und sind verbunden mit den Namen von W. R. Paton, R. Herzog, J. Zarraphtes und M. Segre. Der vorliegende erste Teil des auf fünf Bände geplanten Corpus enthält historisch und religionsgeschichtlich besonders wichtige Texte: 245 Dekrete des koischen Staates und auswärtiger Städte (hervorzuheben sind Ehrendekrete für koische Ärzte aus der berühmten Schule des Hippokrates und zahlreiche Anerkennungsschreiben der Asylie des Asklepieions aus dem Jahre 242 v.Chr.), ferner zahlreiche Briefe hellenistischer Könige und römischer Kaiser sowie 140 Sakralinschriften, die die Kultausübung im Asklepieion und in den zahlreichen Heiligtümern auf der Insel regelten und die Pflichten der Priester vorschrieben. Der zweite Faszikel, der die Namenslisten, die Weih- und Ehreninschriften von Kos umfassen wird, ist in Vorbereitung und wird 2012 erscheinen. Alle Texte wurden an den Steinen überprüft und revidiert. Frühere Lesungen und Ergänzungen sind in einem Kritischen Apparat verzeichnet. Beigegeben sind ferner Beschreibungen der Inschriftträger, kurze Kommentare und Sekundärliteratur. Ausführliche Indizes sowie Fotografien aller erhaltener Inschriften sind für den letzten Faszikel vorgesehen.
