Come assistente di Perugino, realizza la sua prima opera attestata, il Testamento e morte di Mosè, nella Cappella Sistina verso il 1482. La sua carriera, che si estende per quarant'anni, è segnata da numerosi incarichi di confraternite e personalità influenti, portandolo da Roma a Cortona, Firenze e Orvieto. Nella cattedrale di Orvieto, crea tra il 1499 e il 1504 il suo ciclo di affreschi più famoso, dedicato al giudizio universale, testimoniando una grande invenzione spaziale e una notevole maestria tecnica.

Voluta da Giacomo di Simone Filippini, un maggiorente della allora Roccacontrada, la tavola destinata alla cappella gentilizia della famiglia nella chiesa di San Francesco, raffigura la Vergine del latte con quattro Santi ed è stata realizzata nel 1508. Una pala che è fra le opere più schiette del pittore per l'attenuarsi dei contrasti luminosi e cromatici in una tenue luce diffusa. Luca Signorelli, allievo di Piero della Francesca, con esperienze fiorentine nella bottega di Antonio Pollaiolo e Andrea del Castagno, celebrato da Giorgio Vasari nelle sue "Vite", morto a Cortona nel 1523, ha operato anche a Loreto, Fabriano, Matelica, Camerino, Urbino. Nell'antica Rocca Contrada, ora Arcevia, Signorelli dimorò a lungo lasciando straordinari capolavori, alcuni dei quali dispersi. La Collegiata, dove sono molto ben conservati il Polittico di San Medardo e il Battesimo di Cristo, accoglie ora molto dignitosamente la Madonna in trono col Bambino e i santi. Nella chiesa vi sono numerose altre opere d'arte di scultori, pittori, incisori che testimoniano l'importanza e la ricchezza di Rocca Contrada nei secoli trascorsi.
